Research Interests
Metabolism: glycogen; pathways and control of glycogen synthesis; intermediary metabolism; gluconeogenesis; substrates for gluconeogenesis; regulation of carbohydrate metabolism; intestinal glucose absorption and disposition; gut hormones.
Insulin: systemic kinetics; secretion (in vivo); insulin removal and degradation; therapeutics; insulin analogs; insulin action.
General: The above issues in diabetes, cirrhosis, exercise. Pharmacokinetics; bioavailability, mathematical models.
Major Research Activities
Dr. Radziuk's long-term research interests have focused on the pathogenesis of Type II (non-insulin dependent) diabetes mellitus, in particular insulin resistance. Ongoing research is on carbohydrate in intermediary metabolism and insulin dynamics, using in vivo experimental models and applying the technology of mathematical modelling.
Dr. Radziuk has extended his research activities to look at exercise and metabolism. He uses three animal models: the swimming rat, the running rat and the running dog. Exercise is a hypoinsulinemic state and thus exercise and recovery from exercise are ideal situations in which to study muscle metabolism, in particular glycogen and the dynamics of insulin.
He is also investigating the interrelationships of glucose and lactate in the control of glycogen synthesis in the perfused rat in order to elaborate in more detail the specific roles of lactate in the control of glycogen metabolism.
Another area of study is the liver and insulin action: a surgical liver bypass model was developed for these experiments which have shown that physically bypassing the liver decreases peripheral insulin sensitivity to less than 50% of normal. The implications of this result in therapy, physiology, transplantation as well as the pathogenesis of insulin resistance are being explored.
The role of the gut in modulating metabolism and its regulation by pancreatic hormones is being assessed using a variety of gut hormones such as galanin and glucagon-like peptide-1.
